On Debian all you need to control ttyS0 is for the relevant users to be in group dialout. You shouldn't need to chmod ttyS0 (since group dialout has read and write by default), and it is unlikely to stay chmod'ed the way you want, unless you fiddled with deep stuff in the scripts that populate /dev at boot.
