My concern would be how to get updates - the beauty of Debian as a distro is the ease with which you can update it and maintain new versions, my guess is the Knoppix distro (which is Debian based) will have a less well maintained set of repositories, or a set which are focused on building working livecd's. Still you can always track down some Knoppix users and ask how well it works running from hard disk.
